چکیده

Chimeric antigen receptor (CAR)-T cell therapy has impressive efficacy in hematological malignancies, but its application solid tumors remains a challenge. Multiple hurdles associated with the biological and immunological features of currently limit CAR-T cells treatment tumors. Using syngeneic mouse models, we recently reported that engineered to concomitantly produce interleukin (IL)-7 chemokine (C–C motif) ligand 19 (CCL19)-induced potent anti-tumor against through an improved ability migration proliferation even immunosuppressive tumor microenvironment. In this study, for preclinical evaluation preceding clinical application, further explored potential IL-7/CCL19-producing human using models mimic Human anti-mesothelin producing IL-7/CCL19 achieved complete eradication orthotopic pre-established malignant mesothelioma prevented relapse downregulated expression. Moreover, mice patient-derived xenograft mesothelin-positive pancreatic cancers exhibited significant inhibition growth prolonged survival following cells, compared conventional cells. Transfer resulted increase not only also non-CAR-T within tissues expression exhaustion markers, including PD-1 TIGIT, on T Taken together, our current study elucidated exceptional their patients

IL-17 and IL-4 Producing CD8+ T Cells in Tumor Draining Lymph Nodes of Breast Cancer Patients: Positive Association with Tumor Progression

Background: CD8+ cytotoxic T lymphocytes have been recently divided based on their cytokine expression profile. Objective: To evaluate the percentages of CD8+ lymphocytes and their effector subsets including Tc1, Tc2 and Tc17 in the tumor draining lymph nodes (TDLNs) of patients with breast cancer. Methods: Single cell suspensions were obtained from TDLNs of 42 patients with breast cancer. Stai...

Efficacy of the novel camptothecin gimatecan against orthotopic and metastatic human tumor xenograft models.

PURPOSE Gimatecan, a novel oral lipophilic camptothecin characterized by favorable features at molecular/cellular level and by a promising profile of preclinical activity, is currently in clinical phase I/II. The aim of the study was to additionally investigate the therapeutic potential of the drug in human tumor xenografts growing in different organs as models representative of tumor growth in...

Efficacy of tumor-targeting Salmonella typhimurium A1-R in combination with anti-angiogenesis therapy on a pancreatic cancer patient-derived orthotopic xenograft (PDOX) and cell line mouse models

The aim of the present study was to examine the efficacy of tumor-targeting Salmonella typhimurium A1-R treatment following anti-vascular endothelial growth factor (VEGF) therapy on VEGF-positive human pancreatic cancer. A pancreatic cancer patient-derived orthotopic xenograft (PDOX) that was VEGF-positive and an orthotopic VEGF-positive human pancreatic cancer cell line (MiaPaCa-2-GFP) as well...

IL-12 primed CD8+ T-cells possess enhanced persistence and anti-tumor efficacy because of greater IL-7 responsiveness

Ex vivo IL-12 priming of tumor-reactive CD8+ T-cells enhances their persistence and subsequent anti-tumor efficacy upon adoptive cell transfer (ACT) into lymphodepleted mice. Since IL-7 and IL-15 are considered critical for the persistence of adoptively transferred T-cells, we investigated the responsiveness of IL-12 primed CD8+ T-cells to these homeostatic cytokines to help elucidate the mecha...

Anti-GPC3-CAR T Cells Suppress the Growth of Tumor Cells in Patient-Derived Xenografts of Hepatocellular Carcinoma

BACKGROUND The lack of a general clinic-relevant model for human cancer is a major impediment to the acceleration of novel therapeutic approaches for clinical use. We propose to establish and characterize primary human hepatocellular carcinoma (HCC) xenografts that can be used to evaluate the cytotoxicity of adoptive chimeric antigen receptor (CAR) T cells and accelerate the clinical translatio...
